چکیده

There have been several recent electron-microscopic studies on the development of the vascular cambium (Bouck & Cronshaw, 1965; Buvat, 1963, 1964; Cronshaw, 1967; Esau & Cheadle, 1965; Srivastava & O'Brien, 1966; Northcote & Wooding, 1966; Wooding & Northcote, 1964; Wooding, 1966). The organelles involved have been tentatively identified and functions for each suggested. The present report is of a study using electron-microscope radioautography in parallel with a chemical analysis of sycamore vascular tissue, in order to define the functions of the organellar systems involved more closely. Tritiated glucose and phenylalanine were fed to allow a comparison of wall development including lignification.

The Structure of Plant Cell Walls: II. The Hemicellulose of the Walls of Suspension-cultured Sycamore Cells.

The molecular structure, chemical properties, and biological function of the xyloglucan polysaccharide isolated from cell walls of suspension-cultured sycamore (Acer pseudoplatanus) cells are described. The sycamore wall xyloglucan is compared to the extracellular xyloglucan secreted by suspension-cultured sycamore cells into their culture medium and is also compared to the seed "amyloid" xylog...

Immunogold localization of xyloglucan and rhamnogalacturonan I in the cell walls of suspension-cultured sycamore cells.

PLANT CELL WALLS SERVE SEVERAL FUNCTIONS: they impart rigidity to the plant, provide a physical and chemical barrier between the cell and its environment, and regulate the size and shape of each cell. Chemical studies have provided information on the biochemical composition of the plant cell walls as well as detailed knowledge of individual cell wall molecules. In contrast, very little is known...

Radioautographic Study of Cell Wall Deposition in Growing Plant Cells

Segments cut from growing oat coleoptiles and pea stems were fed glucose-(3)H in presence and absence of the growth hormone indoleacetic acid (IAA). By means of electron microscope radioautography it was demonstrated that new cell wall material is deposited both at the wall surface (apposition) and within the preexisting wall structure (internally). Quantitative profiles for the distribution of...

Radioautographic Studies of Choline Incorporation into Peripheral Nerve Myelin

This radioautographic study was designed to localize the cytological sites involved in the incorporation of a lipid precursor into the myelin and the myelin-related cell of the peripheral nervous system. Both myelinating and fully myelinated cultures of rat dorsal root ganglia were exposed to a 30-min pulse of tritiated choline and either fixed immediately or allowed 6 or 48 hr of chase incubat...

Studies on hypoxia: XI. Long-term effects on the epiphyseal plate--a histomeric and radioautographic study.

Rats were exposed to hypoxia for one week. The mean thickness of epiphyseal plates form control rats was 430 micrometers (mum) which was reduced to 313 mum in hypoxic rats. Radioautographic incorporation of 3H-phenylalaine by connective tissue cells in hypoxic rats was reduced up to 38% in control rats.
